Biodegradable scaffolds for enhancing vaccine delivery

Abstract Sustained release of vaccine components is a potential method to boost efficacy compared with traditional bolus injection. Here, we show that biodegradable hyaluronic acid (HA)‐scaffold, termed HA cryogel, mediates sustained antigen and adjuvant in vivo leading durable immune response. Delivery from subcutaneously injected cryogels was assessed formulation which enhanced the response while minimizing inflammation associated foreign body identified, CpG‐OVA‐HAC2. Dose escalation studies CpG‐OVA‐HAC2 demonstrated both antibody T cell responses were dose‐dependent influenced by competency neutrophils perform oxidative burst. In immunodeficient post‐hematopoietic stem transplanted mice, immunization elicited strong response, three orders magnitude higher than dose‐matched melanoma model, induced dose‐responsive prophylactic protection, slowing tumor growth rate enhancing overall survival. Upon rechallenge, none mice developed new tumors suggesting development robust immunological memory long‐lasting protection against repeat infections. also survival established tumors. The results this work support for enhance delivery.
